Genome Regulation in Mycobacterium Tuberculosis


My PhD in the lab of Eric Galburt focused on DNA-dependent processes (transcription and repair) in the pathogenic bacteria Mycobacterium tuberculosis. Using a combination of transient and steady-state kinetics, kinetic modeling, fluorescence anisotropy equilibrium titrations, and analytical ultracentrifugation (AUC), I investigated the regulation of transcription by transcription factors, small molecules, and various sigma factors. By making comparative kinetic measurements to well studied E. coli systems, I highlighted ways in which mycobacteria has diverged evolutionarily and functionally. I am currently continuing aspects of this work in the lab of Andrea Soranno, assessing protein conformational dynamics of the transcriptional machinery using single-molecule FRET.
